Solve Single-Cell Heterogeneity in Disease
Understanding why genetically identical cells behave differently in disease states could reveal new therapeutic approaches. Even cells with identical DNA can respond differently to treatments, making diseases like cancer difficult to cure completely. Single-cell differences in protein expression, metabolism, and stress responses create treatment-resistant populations that can cause disease recurrence. Mapping and understanding this heterogeneity at the molecular level could enable treatments that target all cell variants simultaneously. The challenge involves developing methods to analyze individual cells, identifying sources of variation, predicting cell behavior, and designing therapies that account for heterogeneity. Success could eliminate treatment resistance in cancer, improve outcomes for genetic diseases, and enable more precise therapeutic approaches. This could significantly improve survival rates and reduce disease recurrence across many conditions. Barriers include technical challenges in single-cell analysis, computational complexity of modeling cell populations, understanding sources of heterogeneity, and developing therapies that target multiple cell states simultaneously.
